納米金Gold Nanoparticles的制備方法主要采用還原劑,如檸檬酸鈉,硼氫化鈉等還原氯金酸HAuCl4·4H2O。氯金酸HAuCl4·4H2O在還原劑的作用下,可以聚一定大小的金納米顆粒,形成帶負(fù)電的疏水膠溶液。由于靜電作用而成為穩(wěn)定的膠體狀態(tài),故稱膠體金Colloidal gold。

納米金隨粒徑的變化而顏色發(fā)生變化:不同尺寸形貌的納米金對(duì)可見(jiàn)光有選擇性吸收,吸收了某個(gè)波長(zhǎng)的光后,將沒(méi)有吸收的光反射出去,我們看到的就是反射的光,其實(shí)不是他們的顏色,不過(guò)是他們將這種顏色的光反射給我們看到了,這種光在我們的視覺(jué)中是某種顏色而已。不同尺寸形狀的顆粒,它們的阻尼現(xiàn)象不同,一般來(lái)說(shuō)小顆粒的阻尼現(xiàn)象,從而小顆粒振動(dòng)頻率要小于大顆粒,它們也就吸收長(zhǎng)波的光,反射短波的光。隨著顆粒增加,它們吸收的光波波長(zhǎng)變短,也就是所謂的紅移現(xiàn)象了。

1 納米金應(yīng)用于毛細(xì)管電泳檢測(cè)尿液中8-OHdG

3 納米金應(yīng)用于的研究進(jìn)展

   金納米粒子具有的生物親和性及的表面性質(zhì),經(jīng)表面修飾的金納米粒子通過(guò)共價(jià)或非共價(jià)的方式與靶向分子或者相連,獲得多功能納米粒子,應(yīng)用于的靶向與。金納米粒子是一種的靶向制劑,可以同時(shí)實(shí)現(xiàn)被動(dòng)靶向和主動(dòng)靶向:有納米粒徑大小引起的通透性和滯留效應(yīng)(enhanced permeability and retention effect , EPR )可以實(shí)現(xiàn)被動(dòng)靶向,使納米金載體在組織選擇性分布,以增加并減少副作用。此外,金納米粒子進(jìn)行表面修飾后,可利用靶向分子實(shí)現(xiàn)的主動(dòng)靶向,將或射線特異帶到部位,靶向的目的。納米金團(tuán)簇表面的邊緣位點(diǎn)和頂部位點(diǎn)4f軌道的結(jié)合能相對(duì)較低,這些位點(diǎn)形成的Au-S鍵的鍵能也相對(duì)較低,容易發(fā)生離解,這對(duì)Au-MMPCs的穩(wěn)定性和配體交換有影響.Au-MPCs具有粒徑小和生物相容性好的,用巰基或二硫化合物對(duì)Au-MPCs表面進(jìn)行修飾,形成具有生物活性的Au-MMPCs,這些生物活性Au-MMPCs同待轉(zhuǎn)運(yùn)物和膜相互作用,通過(guò)靜電吸附和內(nèi)吞作用可將轉(zhuǎn)運(yùn)物傳送和核內(nèi),靶向的傳輸和釋放需要依靠Au-MPCs的作用.

4 納米金生物探針及其應(yīng)用

    納米金顆粒的表面不表現(xiàn)為靜電作用,還有疏水作用。為了讓蛋白質(zhì)與納米金顆粒結(jié)合,減小靜電作用,而是疏水作用起作用。通過(guò)表面改性,讓納米金帶正電,從而與帶負(fù)電的蛋白質(zhì)相結(jié)合,制成用于電子顯微鏡的金探針。膠體金與抗人球蛋白(anti human-IgC)結(jié)合制備的金探針可以定性檢測(cè)艾滋(HIV)抗體。在納米金微粒上修飾DNA探針,由于共價(jià)結(jié)合力,在DNA探針的一段連接上巰基后可與納米金結(jié)合,通過(guò)識(shí)別顏色的變化,就能檢測(cè)出相應(yīng)的靶DNA

5納米金在生物標(biāo)記分析中的應(yīng)用進(jìn)展

   金納米粒子除具有一般納米粒子的表面效應(yīng),量子尺寸效應(yīng)外,同時(shí)具有的生物相容性。由于生物大分子和功能化同處于納米維度當(dāng)在相應(yīng)的生物體系中加入功能化的納米金顆粒使其具有定向吸附效應(yīng)以及富集效應(yīng)等。兩者通過(guò)化學(xué)或物理吸附可制得既穩(wěn)定又具有一定計(jì)量關(guān)系的生物/納米復(fù)合材料。該復(fù)合材料又可以用來(lái)構(gòu)建具有功能的分子結(jié)構(gòu)體系,此外也可以作為檢測(cè)探針用于研究復(fù)雜的生物化學(xué)體系。

6 納米金在生物傳感器中的應(yīng)用

   膠體金增DNA生物傳感器的檢測(cè)靈敏度,納米級(jí)的金顆粒標(biāo)記在DNA分子上,可增加該分析分子的質(zhì)量,從而增質(zhì)量性。膠體金能提供一似于能保持蛋白質(zhì)穩(wěn)定性的,實(shí)習(xí)附在其表面的蛋白質(zhì)保持他們的生物活性,能增乳酸生物傳感器的檢測(cè)靈敏度。用金納米粒子修飾鉑電而制成的葡萄糖氧化(GOD)生物傳感器,具有靈敏度,速度快等。
