| I. Background | |
白細胞介素-2 (IL-2)是一種 o 糖基化的四α -螺旋束細胞因子，主要由活化的 T 細胞、 樹突狀細胞和 B 細胞產生。作為一種細胞毒性 T 淋巴細胞（CTL）和 NK 細胞的強效誘導 劑，IL-2 是最早被獲批用于治療轉移性黑色素瘤和腎細胞癌癥的治療藥物之一。但是， IL-2 免疫療法由于其體內半衰期短和治療劑量下的嚴重毒性，因此獲批以后并未得到廣泛 應用。IL-2R 由 IL2Rα /CD25、IL2Rβ /CD122 和 IL2Rγ /CD132 亞基組成。IL-2 與 CD25 的 結合誘導調節(jié)性 T 細胞（Tregs）的增殖，是由于 CD25 會優(yōu)先在 Tregs 上表達，而 Treg 細 胞的耗竭已被證明可以增強 IL-2 誘導的抗腫瘤免疫，這表明 Treg 可能是 IL-2 介導的 CTL 擴增的主要障礙。 | |
