Description:
PeproTech offers three separate endothelial cell culture media formulations developed for the in vitro c*tion of: endothelial progenitor cells (EPC; PeproGrow EPC) derived from bone marrow or peripheral blood; endothelial cells from large vessels (PeproGrow MacroV); and endothelial cells from small vessels (PeproGrow MicroV). These media formulations maintain outstanding endothelial cell morphology and function, and increase the activity of endothelial nitric oxide synthase (eNOS), which account for a specific, crucial marker for endothelial cells. By doing this, the media provide an optimal cell culture environment for macrovascular and microvascular endothelial cells, as well as for EPC; growing cells at rates that exceed commercially available media. These media formulations are optimized for primary/progenitor human cells, but can also be used for bovine, porcine, rabbit, rat, and mouse EPC, or mature endothelial cells.
PeproGrow MacroV is supplied as a 500mL bottle of basal medium and a separate, frozen growth factor component.

The proliferation ability of HUVEC was assessed 7 days after cell c*tion using the XTT assay following the manufacturer’s protocol. The proliferation ability of HUVEC was expressed as the average of fluorescence intensity (O.D.) calculated using a plate reader from two independent assays run in triplicates.

Human umbilical vein endothelial cells (HUVECs) are cells derived from the endothelium of veins from the umbilical cord. They are used as a laboratory model system for the study of the function and pathology of endothelial cells (e.g., angiogenesis).[1] They are used due to their low cost, and simple techniques for isolating them from umbilical cords, which are normally resected after childbirth. HUVECs were first isolated and cultured in vitro in the 1970s by Jaffe and others. [2] HUVECs can be easily made to proliferate in a laboratory setting. Like human umbilical artery endothelial cells they exhibit a cobblestone phenotype when lining vessel walls.
人臍靜脈內(nèi)皮細胞來自臍帶血管的內(nèi)皮細胞。因為價格低和分離技術簡單，人臍靜脈內(nèi)皮細胞通常作為內(nèi)皮細胞的功能和生理研究的實驗模型使用。人臍靜脈內(nèi)皮細胞是在1970年由Jaffe等人*分離使用的。在實驗條件下人臍靜脈內(nèi)皮細胞很容易擴增。

1. 將 15-20cm 長的新生兒臍帶放入無菌的 PBS 溶液中儲存。（注：4℃下zui多貯存 24 小時，室溫下不超過 6 小時，否則廢棄）
2. 用一個鈍頭的針頭扎入臍帶靜脈管中，用無菌的 PBS 溶液沖洗 3-5 次，將污血沖洗干凈為止。
3. 用手術鉗夾緊臍帶下端，加入 15ml 的膠原酶（1mg/ml）室溫下消化 15-20分鐘，并不時上下?lián)u動臍帶。
4. 消化完后，將下端手術鉗松開，消化液流入一個 50 ml 無菌離心管中，用無菌的 PBS 溶液沖洗臍帶 2-3 次。
5. 將收集液離心（2000 轉(zhuǎn)/ 分）3 分鐘。
6. 倒去上清。加培養(yǎng)基。
