CAL-51細(xì)胞CAL-51細(xì)胞培養(yǎng)步驟:
1)復(fù)蘇細(xì)胞:將含有1mL細(xì)胞懸液的凍存管在37℃水浴中迅速搖晃解凍,加入5mL培養(yǎng)基混合均勻。在1000RPM條件下離心5分鐘,棄去上清液,補(bǔ)加4-6mL*培養(yǎng)基后吹勻。然后將所有細(xì)胞懸液加入培養(yǎng)瓶中培養(yǎng)過夜(或?qū)⒓?xì)胞懸液加入6cm皿中),培養(yǎng)過夜。第二天換液并檢查細(xì)胞密度。
2)細(xì)胞傳代:如果細(xì)胞密度達(dá)80%-90%,即可進(jìn)行傳代培養(yǎng)。
1、對于貼壁細(xì)胞,傳代可參考以下方法:
1. 棄去培養(yǎng)上清,用不含鈣、鎂離子的PBS潤洗細(xì)胞1-2次。
2. 加1-2ml消化液(0.25%Trypsin-0.53mM EDTA)于培養(yǎng)瓶中,置于37℃培養(yǎng)箱中消化1-2min,然后在顯微鏡下觀察細(xì)胞消化情況,若細(xì)胞大部分變圓并脫落,迅速拿回操作臺,輕敲幾下培養(yǎng)瓶后加5ml以上含10%血清的*培養(yǎng)基終止消化。
3.輕輕吹打細(xì)胞,*脫落后吸出,在1000RPM條件下離心8-10分鐘,棄去上清液,補(bǔ)加1-2mL培養(yǎng)液后吹勻。
4. 按5-6ml/瓶補(bǔ)加培養(yǎng)液,將細(xì)胞懸液按1:2到1:5的比例分到新的含5-6 ml培養(yǎng)液的新皿中或者瓶中。
PS:若客戶收到2ml小管細(xì)胞,收到細(xì)胞后,用75%酒精噴灑整個管子消毒后放到超凈臺或安全柜內(nèi),嚴(yán)格無菌操作;將小管細(xì)胞轉(zhuǎn)移至T25培養(yǎng)瓶或6cm培養(yǎng)皿,加入5ml左右*培養(yǎng)基混勻,放入培養(yǎng)箱過夜培養(yǎng)后查看細(xì)胞密度:若密度未超過80%,換液繼續(xù)培養(yǎng),視情況傳代或者凍存。若密度超過80%,可直接進(jìn)行傳代(方法同上)。
2、對于懸浮細(xì)胞,傳代可參考以下方法:
方法一:收集細(xì)胞,1000RPM條件下離心8-10分鐘,棄去上清液,補(bǔ)加1-2mL培養(yǎng)液后吹勻,將細(xì)胞懸液按1:2到1:5的比例分到新的含8ml培養(yǎng)基的新皿中或者瓶中。
方法二:可選擇半數(shù)換液方式,棄去半數(shù)培養(yǎng)基后,將剩余細(xì)胞懸起,將細(xì)胞懸液按1:2到1:3的比例分到新的含8ml培養(yǎng)基的新皿中或者瓶中。
3)細(xì)胞凍存:待細(xì)胞生長狀態(tài)良好時,可進(jìn)行細(xì)胞凍存。貼壁細(xì)胞凍存時,棄去培養(yǎng)基后加入少量胰酶,細(xì)胞變圓脫落后,進(jìn)行離心收集,1000RPM條件下離心8-10分鐘,去除上清,按凍存數(shù)量加入血清及DMSO,凍存比例為90%FBS+10%DMSO。
