Operation steps for cell culture

1. 吸走用于培養(yǎng)基，加入3-5mL PBS后輕輕晃動(dòng)培養(yǎng)瓶潤洗細(xì)胞；
2. 吸干凈PBS后加入1mL 0.25%-0.53mM EDTA，輕輕搖晃培養(yǎng)皿使浸沒細(xì)胞表面，培養(yǎng)瓶放37度培養(yǎng)箱消化；
   （細(xì)胞對(duì)于消化比較敏感，消化過度會(huì)嚴(yán)重影響細(xì)胞狀態(tài)，導(dǎo)致細(xì)胞傳代死亡漂浮或者傳代后不長。細(xì)胞消化到細(xì)胞間隙變大但未脫落，可以輕輕吹下時(shí)即可終止，禁止消化到細(xì)胞漂浮?；靹蚣?xì)胞時(shí)盡量輕柔，不要吹出大量氣泡。）
3. 加入6-8ml培養(yǎng)基終止消化，輕輕吹下細(xì)胞混勻；
4. 將混勻的細(xì)胞1000rpm（約150g）離心3min，棄上清，再用新鮮培養(yǎng)基重懸37度培養(yǎng)箱繼續(xù)培養(yǎng)；

傳代比例: 不同細(xì)胞生長速度不一，具體傳代比例視細(xì)胞生長速度而定，大部分細(xì)胞適用1:3-1:4傳代，生長較慢的細(xì)胞可以1:2傳代。
